人胚胎干細(xì)胞和誘導(dǎo)多能干細(xì)胞具有分化成外胚層所有細(xì)胞的能力,包括神經(jīng)干細(xì)胞。人多能干細(xì)胞分化的神經(jīng)干細(xì)胞被用與研究神經(jīng)系統(tǒng)發(fā)展,神經(jīng)系統(tǒng)疾病建模,神經(jīng)毒理學(xué)和藥物篩選。
SMAD信號在神經(jīng)誘導(dǎo)起關(guān)鍵作用。SMAD信號的雙重影響,Noggin和SB431542充分的誘導(dǎo)hPSC*、快速的轉(zhuǎn)變?yōu)樯窠?jīng)細(xì)胞。[1].2010年李文林和他的同事發(fā)現(xiàn)glycogen synthase kinase 3 (GSK3) 和 transforming growth factor (TGF-β)的協(xié)同抑制作用,可有效地使hPSC在7天內(nèi)轉(zhuǎn)化為同質(zhì)神經(jīng)上皮。[2].分化的神經(jīng)干細(xì)胞可以在人白血病抑制因子、GSK3和TGF-β的作用下實現(xiàn)自我更新。神經(jīng)干細(xì)胞有分化為多種神經(jīng)細(xì)胞和神經(jīng)膠質(zhì)亞形的能力,通過特殊的誘導(dǎo)。
基于*神經(jīng)細(xì)胞分化理論,我們開發(fā)了多能干細(xì)胞誘導(dǎo)神經(jīng)干細(xì)胞培養(yǎng)基,該培養(yǎng)基為無血清培養(yǎng)基,在貼壁培養(yǎng)條件下可以高效的使hPSCs轉(zhuǎn)化為NSCs。hPSCs在神經(jīng)誘導(dǎo)培養(yǎng)基下可以轉(zhuǎn)化為同源神經(jīng)上皮。用PSCNIM處理7~10天,90%以上的細(xì)胞表達(dá)神經(jīng)干細(xì)胞標(biāo)記物質(zhì):Nestin 和Sox2;同時多能干細(xì)胞標(biāo)志物Nanog的表達(dá)消失。來源于hPSC的NSCs可以在誘導(dǎo)培養(yǎng)基里培養(yǎng)至少5段(大概24天),能明顯形成花環(huán)狀,并在含有EGF和FGF的培養(yǎng)基擴(kuò)展為神經(jīng)前體細(xì)胞,或誘導(dǎo)分化為神經(jīng)細(xì)胞血統(tǒng)的神經(jīng)元和星形膠質(zhì)細(xì)胞。
